چکیده

Abstract We report a novel glycan array architecture that binds the mannose‐specific binding protein, concanavalin A (ConA), with sub‐femtomolar avidity. new radical photopolymerization developed specifically for this application combines grafted‐from thiol–(meth)acrylate polymerization thiol–ene chemistry to graft glycans growing polymer brushes. The propagation of brushes was studied by carrying out grafted‐to/grafted‐from (GTGFRP) at >400 different conditions using hypersurface photolithography, printing strategy substantially accelerates reaction discovery and optimization on surfaces. effect brush height grafting density mannosides ConA systematically, we found multivalent cooperative account unprecedented sensitivity GTGFRP This study further demonstrates ease which can be tailored an as result advantages photolithography.

Host Tissue and Glycan Binding Specificities of Avian Viral Attachment Proteins Using Novel Avian Tissue Microarrays

The initial interaction between viral attachment proteins and the host cell is a critical determinant for the susceptibility of a host for a particular virus. To increase our understanding of avian pathogens and the susceptibility of poultry species, we developed novel avian tissue microarrays (TMAs). Tissue binding profiles of avian viral attachment proteins were studied by performing histoche...

Probing virus-glycan interactions using glycan microarrays.

Glycan microarrays are surfaces that contain immobilized oligosaccharides or glycoconjugates and have proven useful in probing the interactions between glycan-binding proteins (GBPs) and individual glycans. Such glycan microarrays have been especially important in studying virus-glycan interactions, as most viruses express one or more GBPs important for pathogenesis. For studying interactions o...

Hydrogel glycan microarrays.

The technology of hydrogel microchips manufacturing, which was developed previously for covalent immobilization of DNA and proteins, was applied for the preparation of glycochips and combined glyco/protein chips. Microchips consist of hydrogel drops separated with hydrophobic surface. Spacered amino-saccharides and polyacrylamide glycoconjugates were used for immobilization. Gel elements were a...

Fluorescent labeling agents change binding profiles of glycan-binding proteins.

Interactions of glycan-binding proteins (GBPs) with glycans are essential in cell adhesion, bacterial/viral infection, and cellular signaling pathways. Experimental characterization of these interactions based on glycan microarrays typically involves (1) labeling GBPs directly with fluorescent reagents before incubation with the microarrays, or (2) labeling GBPs with biotin before the incubatio...

Are glycan biosensors an alternative to glycan microarrays?

Complex carbohydrates (glycans) play an important role in nature and study of their interaction with proteins or intact cells can be useful for understanding many physiological and pathological processes. Such interactions have been successfully interrogated in a highly parallel way using glycan microarrays, but this technique has some limitations. Thus, in recent years glycan biosensors in num...
